SLU-PP-332 is a synthetic pan-agonist of the estrogen-related receptors (ERRα, ERRβ, ERRγ) and was tested in mice in a groundbreaking 2023 study from Washington University School of Medicine. The results showed that inactive, untrained mice after SLU-PP-332 administration achieved similar treadmill endurance as trained controls — without additional training. Treated mice ran 70 % longer and 45 % further. Mechanistically the compound activates PGC-1α, induces mitochondrial biogenesis in skeletal muscle, increases oxidative phosphorylation and stimulates expression of endurance markers like myoglobin and slow-twitch fibers. In obese mouse models it accelerated fat metabolism and improved metabolic parameters without diet intervention.
